Images with PfAKAL localization (green), Hoechst DNA staining (blue), differential interference contrast (DIC) were taken on rings a trophozoites, b schizonts, c as well as on stage II, d stage III, e stage IV, f and stage V, g gametocytes, infected erythrocytes. “Bright orange” on DIC images indicates haemozoin crystals. The bars represent 2 μm. In asexual parasites PfAKAL was undetectable in ring stages (a), with expression predominantly in late trophozoites and schizonts. Interestingly, a diffuse subcellular localization of PfAKAL was observed in the parasite cytoplasm and in the nucleus in trophozoite stages, whereas it was mostly nuclear in schizonts (b, c).
