Ookinetes had a reduced capacity to develop into (mature) oocysts. At two days after feeding of mosquitoes the number of young oocysts (stained with anti-P28 antibodies) was reduced by 70-90% when compared to wild type. At day 12 after feeding the oocysts numbers were reduced by 65-85%. Day 12 oocysts were significantly smaller than wild type oocysts and no formation of sporozoites was observed. |

Localization of Plasmodium falciparum c-glutamylcysteine synthetase–green fluorescent protein (PfcGCSGFP) and glutathione synthetase-GFP (PfGS-GFP). P. falciparum GS and gGCS C-terminally tagged with GFP were expressed in P. falciparum D10 erythrocytic stages. Fluorescence was analyzed using an Axioskop-2 mot plus microscope (Zeiss) equipped with a Hamamatsu C4742-95 CCD camera and was shown to be present throughout the cytoplasm of both transfected parasite lines, suggesting that GSH biosynthesis is cytoplasmic.
