Di-cre approach with loxP-intron. Incomplete deletion, an increase in especially antisense transcripts. Likely a substantial growth reduction given failure to obtain clones by limiting dilution.

'by gene knock-down strategy, we showed that the cooperation of RNA exosome catalytic subunit PfDis3 and PfMtr4 cofactor played a major role in ns-ncRNAs degradation'

Imaging data (from Malaria Metabolic Pathways)

Coimmunofluorescence assay of three plasmodial exoribonucleases and a core exosome member, PfRrp45, in ring-stage wild-type 3D7 parasites. The rabbit anti-PfRrp45, anti-PfRNase II, anti-PfRrp6 and mouse anti-PfDis3 antibodies were used in these assays. DAPI, 49,6-diamidino-2-phenylindole. Scale bar, 1 mm. Immunofluorescence assay (IFA) analysis revealed that PfRrp6 and PfRNase II localize as distinct foci at the nuclear periphery and that PfDis3 is localized in the cytoplasm adjacent to the nucleus at the ring stage .Zhang Q, Siegel TN, Martins RM, Wang F, Cao J, Gao Q, Cheng X, Jiang L, Hon CC, Scheidig-Benatar C, Sakamoto H, Turner L, Jensen AT, Claes A, Guizetti J, Malmquist NA, Scherf A.
