Localization of Plasmodium falciparum c-glutamylcysteine synthetase–green fluorescent protein (PfcGCSGFP) and glutathione synthetase-GFP (PfGS-GFP). P. falciparum GS and gGCS C-terminally tagged with GFP were expressed in P. falciparum D10 erythrocytic stages. Fluorescence was analyzed using an Axioskop-2 mot plus microscope (Zeiss) equipped with a Hamamatsu C4742-95 CCD camera and was shown to be present throughout the cytoplasm of both transfected parasite lines, suggesting that GSH biosynthesis is cytoplasmic. No fluorescence is seen in the digestive vacuoles (DV) of the parasites or the erythrocyte host cell (E).
