\"We disrupted the Suf pathway using a dominant negative mutant of SufC and showed that these parasites only survive when cultured in the presence of IPP. Furthermore, these parasites lack the apicoplast organelle and its organellar genome – a phenotype not observed when isoprenoid biosynthesis was specifically inhibited with fosmidomycin. Taken together, these results demonstrate that the Suf pathway has a fundamental role in maintaining the apicoplast organelle in addition to any role in isoprenoid biosynthesis.\" |

Plasmodium falciparum PfSufC is targeted to the apicoplast and may be peripherally membrane-associated. (A) Immunofluorescence localisation of SufC in the Plasmodium falciparum D10 ACPleader-GFP line. SufC signal (red, a), GFP (green, b) and their overlap (c) in two trophozoite examples. Parasite nuclei stained with DAPI (d), and the overall merged figure with the phase contrast scan (e) are shown. The signals obtained for a mitochondrial marker (MitoTracker Deep Red, a), SufC (green, b) and nucleus (blue, d). SufC was not localised to the mitochondrion.Kumar B, Chaubey S, Shah P, Tanveer A, Charan M, Siddiqi MI, Habib S.
