Immunofluorescent imaging of thin blood films of P. falciparum–infected red cells. (A) ring stage, (B, C) trophozoite stages, (D, E) schizont stage parasites. C10 parasites were fixed in cold methanol and acetone. Immunostaining was with anti-Pfmyo-D and parasites were counterstained with DAPI. The DIC and DIC overlay images are also shown. Ring stages did not label. In trophozoites focal staining was found at the parasite’s periphery towards the nucleus. The strongest staining was observed in schizonts; these showed more general cytoplasmic staining than trophozoites with areas of more intense staining neighbouring the nuclei. The IT04 schizont (E) shows punctate fluorescence that appears to be associated with nuclei when visualized in the DAPI overlay.
