P. berghei ANKA Ookinete
Difference from wild-type
RMgm-1241
In vitro: Comparable activation of female gametocytes and production of zygotes, and a slight, but significant decrease in ookinete conversion rates for PbATPKO in comparison with wild type parasites. In vivo (mosquito midgut): The proportion of PbATPKO activated females was substantially reduced in vivo (under 2%) compared with wild type (40%). Similarly, the PbATPKO line generated far fewer zygotes (0%) and ookinetes (0%) in vivo. Failure to activate, generate zygotes, and mature to ookinetes by the PbATPKO female gametocytes in vivo was also evident in the preponderance of aberrant females in vivo (90%) compared with in vitro (1%). No ookinetes were observed at 22 h.

Imaging data (from Malaria Metabolic Pathways)

Localization of epitope-tagged ATP synthase subunits detected using immuno-fluoresence microscopy. Immunofluoresence of alpha, gamma, delta, epsilon, OSCP and c subunits expressed under control of the mRPL2 promoter. Expression of the beta subunit was under the control of its own promoter. Green fluorescence: Anti-HA antibody (AlexaFluor 488-conjugated secondary antibody). Red fluorescence: Mitotracker Red CM-H2XRos. Blue fluorescence: DAPI (4'-6-Diamidino-2-phenylindole). DIC: Differential interference contrast microscopy.Balabaskaran Nina P, Morrisey JM, Ganesan SM, Ke H, Pershing AM, Mather MW, Vaidya AB.
