Imaging data (from Malaria Metabolic Pathways)

Localisation of PHIST:GFP proteins. The left- and right-hand images show GFP localisation and a phase contrast image, respectively. White filled arrows: peripheral GFP puncta; White unfilled arrow: GFP puncta in host erythocyte cytosol. Scale bar, 2 mm. GFP-tagged PF3D7_0401800, PF3D7_0424600, PF3D7_0532400, PF3D7_1102500 and PF3D7_1476200 were all exported to the host cell and displayed a striking localisation at the edge of the host erythrocyte (1A–E), PF3D7 1252700 was clearly peripheral in the host erythrocyte (F). PF3D7_0201600 (G) shows weak accumulation at the erythrocyte periphery, but the majority of the protein was localised in the RBC cytosol. PF3D7_0936600 was localised in the erythrocyte cytosol, not at the host cell periphery (H).Tarr SJ, Moon RW, Hardege I, Osborne AR. A conserved domain targets exported PHISTb family proteins to the periphery of Plasmodium infected erythrocytes. Mol Biochem Parasitol. 2014 196(1):29-40 PMID: 25106850

Time course analysis of PfGEXP5 expression. (b) Representative immunofluorescence images of (top panel) a ring stage-like parasite at 14 h pi positive for PfGEXP5 (green) and negative for Pfg27 (red), and (lower panel) a pigmented double-positive parasite at 38 h pi. (c) Immunofluorescence microscopy of ~18-h gametocyte and asexaul ring-infected RBCs labelled with anti-PfGEXP5 (green) and anti-RESA (red) antibodies. Brightfield images (BF) and nuclear staining are shown at right and left, respectively. Scale bar 5 μm.Tibúrcio M, Dixon MW, Looker O, Younis SY, Tilley L, Alano P. Specific expression and export of the Plasmodium falciparum Gametocyte EXported Protein-5 marks the gametocyte ring stage. Malar J. 2015 14:334. PMID:

c Immunofluorescence microscopy of sexually induced PfGEXP5-GFP transfectants (14 h pi), probed with rabbit anti-GFP (green) and mouse anti-Pfs16 (red) antibodies. PfGEXP5-GFP is expressed and exported to the host cell cytoplasm in Pfs16-positive cells (c) d Immuno-fluorescence microscopy of sexually induced PfGEXP5-GFP transfectants (14 h pi, top) and an asexual trophozoite from the same line (bottom), probed with anti-GFP (green) and anti-KAHRP (red) antibodies. Scale bars 3 μm. PfGEXP5-GFP positive cells did not exhibit KAHRP labelling at the RBC surface, while KAHRP-positive cells (i.e., asexual stages) were negative for exported PfGEXP5-GFP (d).Tibúrcio M, Dixon MW, Looker O, Younis SY, Tilley L, Alano P. Specific expression and export of the Plasmodium falciparum Gametocyte EXported Protein-5 marks the gametocyte ring stage. Malar J. 2015 14:334.
